چکیده
α-Glucosidase inhibitors are effective in controlling postprandial hyperglycemia, which play crucial roles the management of type 2 diabetes. Protocatechuic acid (PCA) is one phenolic acids existing not only various plant foods but also as a major microbial metabolite dietary anthocyanins large colon. The present study investigated inhibitory mechanism PCA on α-glucosidase in vitro and examined its effect blood glucose levels vivo. Results from experiments demonstrated that was mix-type inhibitor α-glucosidase. Driven by hydrogen bonds van der Waals interactions, reversibly bound with to form stable α-glucosidase-PCA complex spontaneous manner. computational simulation found could insert into active cavity establish catalytic amino residues. binding aroused steric hindrance for substrates enter sites caused structural changes interacted exhibited hypoglycemic capacity diabetic mice. This may provide theoretical basis application an ingredient functional

Recent studies, including our animal experiments, indicate that this simple phenolic acid could be protective against the development of epithelial malignancy in different tissues and cardio...
متن کاملCholesterol metabolism in rat is affected by protocatechuic acid.
The effects of protocatechuic acid on serum cholesterol and gene expression related to cholesterol metabolism in rats were investigated. Rats were fed a cholesterol-free diet with or without 5 g protocatechuic acid/kg diet for 4 wk. There were no significant differences in body weight and food intake among groups through the experimental period. The liver weight in the protocatechuic acid group...
متن کاملThe metabolism of protocatechuic acid by Neurospora.
A mutant strain of Neurospora crassa, Y7655a, which requires for growth either phenylalanine, tyrosine, tryptophan, and p-aminobenzoic acid in combination or shikimic acid alone, was found to be blocked in the conversion of dehydroshikimic acid t.o shikimic acid. Some dehydroshikimic acid was accumulated during growth, but the major accumulated product was protocat.echuic acid (3,4-dihydroxyben...
متن کاملInvolvement of oxidative stress in protocatechuic acid‐mediated bacterial lethality
The involvement of oxidative stress in protocatechuic acid-mediated bacterial lethality was investigated. Minimum inhibitory concentrations (MIC) and minimum bactericidal concentration (MBC) of protocatechuic acid against Escherichia coli, Pseudomonas aeruginosa, and Staphylococcus aureus are 600 and 700 μg/ml, 600 and 800 μg/ml, and 600 and 800 μg/ml, respectively. The optical densities and co...
